Where these five locks came from

None of these medicines arrived without a lab, a trial, and a regulator who set the edge of the claim.


Pfizer Sandwich (former discovery site)

Sildenafil began life here as compound UK-92,480, tested first as a treatment for angina. The heart data underwhelmed; a side effect the trial volunteers kept mentioning did not, and the programme was redirected. Viagra, and with it the whole PDE5 class, traces back to this Kent campus.

Tadalafil was identified at this Washington biotech and carried through late-stage trials in partnership with Eli Lilly. Its defining trait was pharmacokinetic rather than chemical — it cleared the body far more slowly than its rivals, which is the single fact that turned it into the long-window tablet the category now benchmarks against.

Bayer AG, Wuppertal

The Wuppertal laboratories produced vardenafil, later marketed as Levitra and the orodispersible Staxyn. Milligram for milligram it binds PDE5 more tightly than either sibling, which is why its labelled doses read so much smaller than sildenafil's.

Ciprofloxacin came out of Bayer's fluoroquinolone programme in the early 1980s — a fluorine atom and a piperazine ring bolted onto the older nalidixic-acid scaffold, producing a broad-spectrum antibiotic that reaches tissues oral drugs usually cannot. The same chemistry explains both its reach and the tendon and nerve cautions now printed in bold on its label.

Parke-Davis / Warner-Lambert (historic)

Gabapentin was designed at Parke-Davis as a GABA analogue meant to calm over-firing neurons; it never touched GABA receptors as intended, but it damped calcium channels instead and found a durable role in nerve pain and epilepsy. Warner-Lambert, and later Pfizer, took Neurontin from this Michigan research base to market.

The American regulator whose approved prescribing information fixes the doses, boxed warnings and interaction rules we quote. Where a page here and the current FDA label diverge, the label is the authority — the nitrate contraindication for the PDE5 drugs and the fluoroquinolone boxed warning above all.

Why the map is here

Sildenafil started as an angina candidate at Sandwich and kept a cardiovascular caution after the programme turned. Tadalafil's slow clearance out of Bothell is why a once-daily 5 mg tablet exists at all. Vardenafil's tighter PDE5 binding out of Wuppertal is why 2.5 mg is a real labelled start. Ciprofloxacin's fluorine-and-piperazine reach out of Leverkusen is why the boxed warning is long. Gabapentin missed GABA receptors in Ann Arbor and still found a role on calcium channels. Each profile points back to the institution that shaped the molecule.
